PU - Jiangsu Academy of Agricultural Sciences LP - Nanjing CP - China AB - A fast and efficient method for isolation of total RNA from leaves of strawberry (Fragaria spp.) in vitro and field was developed. The RNA samples of the leaves of strawberry were isolated by improved CTAB method, and the purity and quantity of each RNA obtained was evaluated by agarose gel electrophoresis, UV scanning and RT-PCR analysis. The total RNA of strawberry leaves extracted by this method showed clear hands of 28 S rRNA, 18 S rRNA and 5.8 S rRNA, and the values of OD260/OD280 were between 1.83 and 2.14. The cDNA reversely transcripted from the extracted RNA exhibited clear band by RT-PCR, which meant that the quality and purity of the RNA could meet the demands of molecular biology experiment. The results showed that H5-H9 bivalent inactivated vaccine was effective in the prevention of avian influenza in Wuxi district, China. Enhancing the detection of infection ratio in waterfowl and birds is the basis of preventing the virus from the flocks. It indicated that the immune procedure may he helpful for efficient control of the disease. PU - Jiangsu Academy of Agricultural Sciences LP - Nanjing CP - China AB - To study the heredity pattern and identify the mutation gene of a kind of white spot mouse (named Wbct) obtained by ethylnitrosourea (ENU) mutagenesis, wbct heterozygotes mice were mated with normal C57BL/6 (B6) mice. Their offspring with normal or mutation phenotype were registered respectively and x2 test was used to confirm the heredity pattern. Fine mapping was conducted by increasing (B6 x D2) x B6, N2 mice DNA sample and microsatellite marker. Based on fine mapping, the candidate gene Pax3 was found. Pax3 gene from wbct heterozygote gene was amplified by PCR followed by sequencing. Heredity test indicated that the white spot phenotype is controlled by a single dominant gene with 100% penetrance but was affected by genetic background. The homozygous mice could not survive. A c-to-a transversion at the 264th nucleotide in WbctM ORF introduced a pre-termination codon of amino acid and caused the phenotype al dominant white spot. The results showed that only BB genotype distributed in detected sheep populations, however, three kinds of genotypes (AA, AB, and BB) distributed in goat populations. In Boer goat and the Yangtze River delta white goat, the distribution of AA genotype was highest, that of AB genotype was in the middle, and that of BB genotype was use lowest. In Huanghuai goat, the distribution of all genotype was the highest, that of BB genotype was in the middle, and that of AA genotype was the lowest. These results afforded a good foundation for further study on the influence of MSTN gene on the growth trait of goats. In addition to 17, 24, 32, 33, and 34, the other 29 serotypes were positive. Homology analysis showed nucleotide sequence homology were 88.3%-100.0% and the derived protein sequence homology were 94.7%-100.0%. It provides opportunities for further study on biological characteristics and functions of IMPDH. It was found that the NH4+-N and NO3--N in the soil profiles gradually decreased from surface soil layer to bottom soil layer. NH4+-N content was stable below the surface soil. In Whitish soil, there was increasing accumulation of available P with increase in soil depth, which reached the maximum amount at a depth of 12-20 cm. In the Wushan soil, available P decreased with soil depth. The two soils showed little difference in the pH value. PU - Jiangsu Academy of Agricultural Sciences LP - Nanjing CP - China AB - Field experiment were established to study the effect of mechanized straw application on the yields of rice and wheat and soil labile carbon content variation in Tai lake region in China. The plant height, shoot and root biomass of wheat seedlings on average were increased by 5, 16 and 13.0%, respectively, after 3 years of continuous application of straw. Rice and wheat yields were 2.1-8.6% and 5.1-13.0%, respectively, higher in straw-applied fields than in the field with no application of straw (control). Soil organic matter (OM) content was increased and each OM ingredient reacted discrepantly to straw application. Labile OM increased the most, being 2.5 times higher than the initial soil, while the total OM increased the least, the increasing rate was 26% only. Compared with carbon management index (CMI) of the initial soil, the soil CMI of half-amount and total-amount of straw application was increased by 1.52-1.94 and 1.22-1.99, respectively. Compared to the control, the soil CMI of half-amount and total-amount of straw application was increased by 67.7 and 72.0%, respectively, indicating total-amount application of straw was more effective in improving the soil CPMI. The soil CMI in rice season was significantly lower than that of wheat season. Analysis on the correlation between crop yields and soil OM ingredients suggested that the labile OM and CMI were better related to crop yield than total OM. Consequently, the soil labile OM and CMI are better evaluation indexes than the total OM in view of soil productivity.
